ABSTRACT

Pancreatic ductal adenocarcinoma (PDAC) is a largely incurable disease, with limited options for early diagnosis and therapy. Technological advancement such as high-resolution spatial multi-omics and the advanced machine learning (ML)-based analysis of human pre-neoplastic and tumor tissues have the potential to reveal prognostic biomarkers and novel therapeutic targets for PDAC. Concurrently, the development of innovative cell and gene therapies based on gene engineering of adaptive and innate immune cells represent a major goal for PDAC treatment. Here, we will combine state-of-the-art technologies for single-cell spatial transcript and protein expression in primary human samples with ML-powered tissue image analysis tools to elucidate the spatial and molecular organization of the immune tumor microenvironment (TME) in PDAC and pre-neoplastic lesions. These data will be correlated with histomorphological heterogeneity, gene expression profiles and clinical parameters to build tissue-based predictive models of disease evolution and to identify novel therapeutic targets informing the design and development of innovative cellular therapies. The unique and leading expertise of the Applicant and Partner in the fields of cell and gene therapy for immune oncology (PI), advanced molecular analysis of the pancreatic TME, access to clinical samples of and pre-neoplastic lesions PDAC, and computational tools for ML-based tissue analysis should provide the highest chances of success.
