Background: Glioblastoma (GBM) is the most aggressive brain tumor with a median survival under two years. Its heterogeneity challenges personalized medicine (PM), as consistent and actionable targets are difficult to identify. Advanced sequencing has provided limited therapeutic leads, and targeted therapies benefit only a minority of patients who often develop resistance.
Objectives: To enhance PM in GBM by developing an Advanced Drug Screening (ADS) platform that integrates patient-derived models, multi-omics data, liquid biopsy and artificial intelligence (AI) to predict drug responses and guide personalized treatments.
Methods: We will recruit treatment-naïve GBM. A library of FDA-approved or clinically developed drugs will be screened in in vitro 3D models, microfluidic organ-on-chip devices, and in vivo patient-derived xenografts (PDX). Plasma extracellular vesicle (EV)-based biomarkers will be used for real-time monitoring of treatment efficacy. Comprehensive multi-omics profiling—including genomic, transcriptomic, and epigenetic analyses—will be performed. An AI-based platform will integrate these data to predict drug efficacy and support clinical decision-making.
Expected Outcomes: By integrating advanced drug screening, multi-omics assessments, liquid biopsy, and AI, this project addresses the challenges of GBM heterogeneity, aiming to enhance PM and contribute to better patient care.
